タイトル:ICP-MS/MSを用いた構造異性体の選択的イオン化

要旨:異性体は同一の分子式を有しているが、構造が異なるため物理的・化学的性質や生体機能・毒性が大きく変化する。このため、化学、生物学、薬学、医学、環境科学、食品科学など幅広い分野において異性体の識別は重要な分析要請となっている。質量分析法は高感度かつ多成分混合物中の目的分子の識別に優れた手法であり、複雑な試料中の分析に広く用いられている。しかし、その識別がm/zに基づく以上、同一分子式を有する構造異性体の判別は依然として課題が残されている。この制約を克服する手段として、ICP-MS/MSのコリジョン・リアクションセル(CRC)内におけるイオン反応に着目した。CRCではICPで生成された一次イオンと揮発性有機化合物との間で電荷交換反応や金属付加反応(cationization)が進行し、標的分子をソフトにイオン化することができる。分子構造はこれらの反応を支配する要因の一つであり、異性体間で反応生成物やその強度に差が生じ得るため、構造選択的なイオン化(異性体識別)への利用が期待できる。

本研究では構造異性体のテスト化合物として1-ブテンおよび2-ブテン(cis?trans混合)を用い、計46種の元素イオンを反応イオンとしてCRC内でのイオン化挙動を比較した。多くの元素イオンでは両異性体に対する反応挙動に顕著な差は認められなかったが、Zn?では両者で明確に異なる挙動が観測された。Zn?は1-ブテンに対してはカチオン付加分子[Zn+C?H?]?を形成したのに対し、2-ブテンでは付加体の生成はほとんど認められず、主として電荷交換反応による分子イオン[C?H?]?が検出された。以上の結果から、ICP-MS/MSのCRC内イオン反応を利用することで、構造異性体に対する選択的なイオン化が可能であることが示唆された。

Title: Molecular ICP-MS: New analytical technique for Volatile Organic Compounds Through Soft Ionisation

Abstract: Volatile organic compounds (VOCs) are a group of various organic chemicals that can have significant effects on both human health and the environment. Some VOCs are harmful and pose risks to living organisms, while others can function as biomarkers to provide insight into disease progression and diagnosis. As a result, the development of new analytical techniques for the rapid and sensitive detection of VOCs has become a critical focus in various research fields such as forensic, environmental, or material sciences. Mass spectrometry is a powerful and user-friendly method for the detection of various molecules and elements. Gas chromatography mass spectrometry (GC-MS), utilizing electron ionisation (EI), is the current gold standard in the detection of VOCs. This technique provides sensitive and quantitative data for individual components from complex mixture of VOCs. The consistent and reproducible fragmentation patterns produced by EI, along with the extensive mass spectral libraries, enable GC-MS to be used for reliable compound identification. However, due to the relatively long time required for a single analysis and labour-intensive sample preparation, this technique is better suited to quantitative analysis rather than qualitative or survey analyses.

In this study, we describe a novel analytical technique using inductively coupled plasma-tandem mass spectrometry (ICP-MS/MS) for detection of organic compounds to overcome the above challenges in VOC analysis. VOCs were introduced directly into the collision/reaction cell (CRC), instead of through the ICP ion source (Fig. 1), and the VOCs were ionised either by protonation or cationisation using metal ions such as Li+, Na+, or Ag+. When ionised with Ar+, ions were detected as protonated ions ([M+H]+), molecular ions (M+), deprotonated ions ([M−H]+), and fragment ions, depending on the VOCs analysed. In contrast, when cationisation using Li+, Na+ or Ag+ was applied, the major product ions were metal-adduct ions (i.e., [M+Li]+, [M+Na]+, or [M+Ag]+), with significantly reduced fragmentation. Additionally, laser ablation was also coupled with the present ICP-MS/MS technique for rapid analysis of VOCs from solid samples. Detailed instrumental design, system setup, and experimental results concerning the simultaneous detection of both elements and molecules will be presented.
