タイトル: LA-ICP-TOF-MS法による生体内微量元素とナノ粒子の同時イメージング分析

Title:Simultaneous imaging analysis of trace elements and nanoparticles for biological samples using a laser ablation-ICP-mass spectrometry

ナノ粒子は,従来の材料と異なる特性を示す物質として注目されており,世界規模で研究がされている.一方で,ナノ粒子による生体や環境への悪影響の懸念もあり,ナノ粒子の毒性は粒径や元素組成などに依存している(Pan et al., 2007 and Cho et al., 2018).また,ナノ粒子由来の溶存イオンも毒性を示す(Wagner et al., 2010).そのため,生体内におけるナノ粒子・溶存イオンの挙動を定量的に評価する目的で,ナノ粒子の粒径,溶存イオンの濃度,分布情報を同時に取得可能な分析技術が求められている.また,生命活動は様々な化学反応と生体分子の相互作用が複雑に進行している.特に,金属元素は遺伝子発現や代謝反応に関わるタンパク質に含まれ,金属元素の化学形態や含有量により生理機能に大きな影響を与える(Haraguchi et al., 2004).こうした背景から,1つの元素・生体分子に注目した一義的な理解では詳細な毒性メカニズム・生命現象の解明において不十分であり,ナノ粒子や生体内金属元素を網羅的に定量・分布分析を行う必要がある.

固体試料中の元素分析法としてレーザーアブレーション法(LA法)と誘導結合プラズマ質量分析法(ICP-MS法)を組み合わせたLA-ICP-MS法がある(Becker et al., 2014).LA-ICP-MS法では固体試料の局所的な元素分析が可能であり,さらに局所分析の二次元平面への展開により元素分布情報も得られる.近年では,LA-ICP-MS法をナノ粒子分析に応用したLA-spICP-MS法も報告されている(Yamashita et al., 2019).しかしながら,先行研究では磁場型質量分析計を用いているため単一同位体(元素)しか計測できていない.そこで本研究では,多元素同時分析が可能である飛行時間型ICP-MSに着目した.

また,ナノ粒子に由来する溶存イオン濃度や生体内金属元素濃度の同時定量分析の報告例はない.LA-ICP-MS法による元素濃度の定量分析には,分析試料と主成分が一致した標準物質が必要である.主成分が異なる場合,アブレーション効率や非スペクトル干渉が原因で確度の低い結果が得られてしまう.一般的に,標準物質にはアメリカ国立標準技術研究所が作製した標準ガラス試料が用いられているが,主成分が二酸化ケイ素であるため炭素を主成分とした生体試料の定量分析には適していない.以上より,本研究ではナノ粒子の粒径分析と,溶存イオン・生体内金属元素濃度の定量分析を可能とする標準物質の作製と,多元素同時定量イメージング分析法の開発を目指す.本発表では分析法や分析条件の検討を説明するとともに,現状で得られているイメージング結果と課題を紹介する.

タイトル:レーザーアブレーション試料導入法による有機質量分析法の開発

 Imaging of molecular substances using mass spectrometry is a powerful tool to obtain distribution information in solid samples such as chemical, biological, industrial and geological samples. In this regard, much of the progress has been made in detecting macromolecules such as proteins, polymers and nucleic acids rather than small molecules. The most well-established methods include matrix-assisted laser desorption/ionisation (MALDI), secondary ion mass spectrometry (SIMS), and desorption electrospray ionisation (DESI) have been successfully used in mass spectrometry imaging (Eccles & Vickerman, 1989; Caprioli et al., 1997; Cooks et al., 2004). Each approach has a specific sample preparation procedure that if conducted improperly could dramatically impact the amount of usable data that can be required. Until recently, the mapping of small molecule distributions was not easily achieved by these methods for a variety of reasons (Brown et al., 2015). Imaging by MALDI is challenged by high background contributions from the matrix, which results in significant interference in the low mass range of the mass spectrum. In DESI, the need for a solvent can be problematic in cases where the sample is sensitive to surface wetting. Furthermore, the requirements of high vacuum (SIMS, MALDI) can severely limit the range of sample types that can be analysed.  Although these techniques are extremely useful for high-mass measurements, the need for a technique to routinely map the special distributions of small molecules remains.

 Hence, we aim to develop an analytical technique for the imaging of molecules with mass spectrometry that can solve the aforementioned problems, which includes characteristics of analysing the sample under atmospheric pressure conditions with minimal to no sample pretreatment, no need for solvent, routinely detect a range of small molecules, and easy set up. In this study, a 266 nm Nd:YAG laser (MOPA 266-200mW, CryLaS, Berlin, Germany)  was used as the LA sampling device. Organic compounds from solid samples were ablated with LA, and was ionised with an in-house atmospheric pressure plasma-based ion source. The ions generated were then detected with a quadrupole mass spectrometer (QTRAP 5500, AB Sciex, California, USA). This presentation will be reporting on the construction of the LA-atmospheric pressure ionisation system, and the results of organic compounds detected by this system.
